Generation of cell lines using Epstein-Barr Virus (EBV) transformation of small volumes of cryo-preserved whole blood and the use of bench-top flow cytometry to achieve high and reproducible success

چکیده

ECACC has developed a procedure for the generation of lymphoblastoid cell lines (LCLs) by the direct EBV transformation of small amounts (800 l) of cryo-preserved whole blood eliminating any requirement for prior separation of peripheral blood lymphocytes (PBLs). The ability to directly transform frozen whole blood avoids the cost of separating and storing PBLs while retaining the option to make a cell line at any time from all or a subset of the collection. Consistent transformation success rates of over 90% have been achieved using bench-top flow cytometry to track lymphoblastoid proliferation from source blood that has been cryo-preserved for more than four years.

THE IN VITRO GROWTH PROPERTIES OF CELL LINES FROM EPSTEIN-BARR VIRUS-INDUCED TAMARIN TUMORS AND TAMARIN B CELLS TR ANSFORMED BY EPSTEIN BARR VIRUS

EBV-carrying human cell lines, depending on whether the cells are derived from Burkitt's lymphoma (BL) tumor biopsies or transformed by EBV in vitro, have different growth properties in vitro. In contrast, there are no clear differences between tamarin tumor lines and tamarin LCLs in vitro. Both types of tamarin cell lines could grow in agarose and formed colonies unlike human LCLs, althoug...

EBV Immortalization of human B lymphocytes separated from small volumes of cryo-preserved whole blood.

EBV immortalized B lymphocyte cell lines have been extensively used as a source of biological material for functional and molecular studies and represent a potentially limitless source of genomic DNA. Current technologies for EBV transformation are costly and use relatively large volumes of peripheral blood. Alternative methods were examined to determine whether smaller volumes of cryo-preserve...

Immunophenotyping of Nodal Peripheral T-cell Lymphomas and its Association with Epstein-Barr Virus

Background: Immunophenotyping in the rare group of nodal Peripheral T-cell Lymphomas (PTCL) exposes interesting features such as T-cell marker downregulation and paradoxically, the presence of reactive, clustered large-sized CD20 positive B-cells (B-cell proliferation). Epstein-Barr virus (EBV) has been suggested as a putative etiology in pathogenesis of B-cell lymphoma. We aimed to review the ...

Existence of Epstein-Barr Virus in the Umbilical Cord and Sera Samples of Healthy Pregnant Women

Background: Epstein-Barr virus (EBV)-associated post-transplant lymphoproliferative disorder (EBV-PTLD) is a known and serious condition for the hematopoietic stem cell transplantation. This study evaluated the prevalence of EBV in umbilical cord samples of healthy pregnant women in Baghdad, Iraq.  Methods: 800 umbilical cord blood (UCB) samples were collected from healthy pregnant women. The ...

Analysis of Epstein Barr Virus Genome in Serum and Ocular Samples of Patients with Inflammatory Eye Disease Using PCR Method

Background and Aims: Epstein-Barr virus (EBV) infection is very common in the population. Virus belongs to the family Herpesviridae, whose representatives are characterized by the ability to cause the human body latent persistent infections. The goal of this study was to assess EBV infection frequency using PCR method in samples from inflammatory eye disease, in comparison with EBV presence in ...
